does not seem to be a problem - though I only know how to achieve that working with Linux. Should definitely be possible on Windows or OSX. Have a look at with_fx :sound_out_stereo and the following posting:

For those, who are interested: Another proof of concept: A running configuration with Sonic Pi, Nonmixer and Carla with Calf Reverb Plugin applied to one of the live_loops: [sp-nonmixer-carla] Now obviously this quickly becomes a performance issue. I do need same faster hardware …
